Abstract

Cilj ovog rada je dati osvrt na dosadašnja saznanja o ulozi mismatch repair gena (MMR) u jednom od najsmrtonosnijih tumora mozga - multiformnom glioblastomu (GBM). Do sada je poznato kako manjak aktivnosti MMR gena može uzrokovati nastanak tumora, te da je mikrosatelitna nestabilnost u većini slučajeva vezana uz mutacije na MMR genima, no nije otkrivena kod svih tumora. Nadalje, u vezu su stavljeni utjecaji ekspresije MGMT gena i gubitka MMR gena na pojavu recidiva GBM-a kao i na pojavu rezistencije na temozolomid. Naposljetku, dobiven je uvid u nova istraživanja o mogućim primjenama MMR gena u liječenju glioblastoma, a uključuju koncepte sintetičke letalnosti i primjene genske terapije.

Abstract (english)

The aim of this work was to give a review of on record findings of the role of mismatch repair genes (MMR) in the most lethal tumor known - the glioblastoma multiforme (GBM). It is known that the MMR deficiency can cause tumor growth and that it is partly related to the occurrence of the microsatellite instability, but the same phenomenon is not discovered in all tumor samples. Furthermore, the role of MGMT expression as well as MMR deficiency has been investigated in order to find out the mechanisms of GBM recurrence and also development of temozolomide resistance. Ultimately, we have got an insight of possible MMR applications in treating GBM, which includes the concept of synthetic lethality and also application of gene therapy.
