Abstract

Kulture humanih stanica se koriste u industrijskoj biotehnologiji te u brojnim znanstvenim disciplinama, izmeĎu ostalog i kao test sustavi u cilju ispitivanja toksičnosti ksenobiotika. U farmaceutskoj industriji njihova primjena vrlo je vaţna pri proizvodnji visokovrijednih pripravaka poput humanih i veterinarskih cijepiva, hormona, neuroregulatora i sl. Uključivanje in vitro metoda u toksikološka ispitivanja nuţno je u cilju smanjenja broja ţivotinja u pokusima. Osim toga, primjena kulture stanica omogućuje istraţivanja molekularnih mehanizama toksičnosti, interakcija s receptorima i praćenje specifičnih unutarstaničnih promjena izazvanih djelovanjem toksikanta, dok in vivo studije ne omogućuju takve spoznaje. Većina alternativnih testova toksičnosti još je u postupku razvoja i validacije te se koriste samo kao predtestovi. U ovom radu preliminarno je ispitana mogućnost korištenja humanih keratinocita - HaCaT stanične linije kao alternativnog test sustava u ispitivanju potencijalno štetnih tvari kojima su impregnirane tekstilije. Upotreba kulture HaCaT stanica omogućila je vrlo brzo, rutinski i relativno jeftino dobivanje podataka čime se test sustav pokazao zadovoljavajućim.

Abstract (english)

Human cell cultures are used in industrial biotechnology and in numerous scientific disciplines, including as test systems for toxicity testing of xenobiotics. In the pharmaceutical industry, their application is very important in producing high-valuable preparations such as human and veterinary vaccines, hormones, neuroregulators etc. Inclusion of in vitro methods in toxicity studies is necessary - primarily in order to reduce the number of animals used in experiments. In addition, the use of cell cultures allows investigation of molecular mechanisms of toxicity, interaction with receptors and monitoring of specific intracellular changes caused by the activity of toxicant, whereas in vivo studies do not provide such insights. The majority of alternative toxicity tests are still in the process of development and validation and are used only as preliminary tests. In this study, it has been investigated the possibility of HaCaT cell line (human keratinocytes) usage as an alternative test-system in determination of the potential harmfulness of selected textile materials. The usage of HaCaT cell culture proved to be a suitable model to follow toxicity of selected textile. The data were obtained very quickly, routinely and testing procedure was relatively non expensive and reproducible.
