Abstract

Cilj ovog istraživanja bio je odrediti masene udjele klorofila a, klorofila b i karotenoida u ekstraktima liofilizirane smeđe alge Cystoseira proizvedenim pomoću ubrzane ekstrakcije otapalima pri povišenom tlaku (PLE). Uz etanol kao ekstrakcijsko otapalo, ispitivani su sljedeći parametri ekstrakcije: (i) statičko vrijeme ekstrakcije (5, 10 i 15 min); (ii) broj ciklusa ekstrakcije (1 i 2); te (iii) temperatura (100 °C, 130 °C, 160 °C). Maseni udjeli navedenih pigmenata određeni su spektrofotometrijski, a utjecaji parametara ekstrakcije definirani su na temelju provedene statističke analize MANOVA. Maseni udjeli klorofila a određeni su u rasponu od 0,86 do 1,92 mg g-1 s.t., klorofila b od 0,09 do 1,19 mg g-1 s.t. te karotenoida od 0,36 do 0,70 mg g-1 s.t. Utvrđena je statistički značajna razlika u masenim udjelima svih pigmenata u ovisnosti o svim primjenjenim parametrima ekstrakcije, osim kod klorofila b, gdje broj ciklusa ekstrakcije nije statistički značajan parametar. Rezultati istraživanja pokazali su da se najveći prinosi pigmenata dobivaju pri slijedećim uvjetima ekstrakcije: klorofil a pri temperaturi ekstrakcije od 160 °C i jedan ciklus od 5 minuta; klorofil b i karotenoidi pri temperaturi ekstrakcije od 130 °C te jedan ciklus od 15 minuta.

Abstract (english)

The aim of this study was to determine mass fractions of chlorophyll a, chlorophyll b and carotenoids in extracts of lyophilized brown algae Cystoseira, produced by pressurized liquid extraction (PLE). Ethanol was the extraction solvent and the following extraction parameters were investigated: (i) static extraction time (5, 10 and 15 min); (ii) number of extraction cycles (1 and 2); and (iii) temperature (100 °C, 130 °C, 160 °C). Mass fractions of certain pigmentsin obtained extracts were determined spectrophotometrically and the effect of extraction parameters was defined by statistical analysis MANOVA. Mass fractions of chlorophyll a were ranged from 0.86 to 1.92 mg g-1 s.w., chlorophyll b from 0.09 to 1.19 mg g-1 s.w. and carotenoids from 0.36 to 0.70 mg g-1 s.w. A statistically significant difference was determined depending on all applied extraction parameters, except for chlorophyll b, where the number of extraction cycles was not a statistically significant parameter. The results of the research have shown that the highest pigment yields are obtained under the following extraction conditions: chlorophyll a at extraction temperature of 160 °C and one 5 minute cycle; chlorophyll b and carotenoids at extraction temperature of 130 °C and one 15 minutes cycle.
