Utjecaj prirodnih eutektičkih otapala na lipazom kataliziranu hidrolizu (R, S)-1-feniletil-acetata

Abstract

Prirodna eutektička otapala mogu se primijeniti u biotransformacijama za proizvodnju biološki aktivnih kiralnih spojeva ili kiralnih sintona visoke enantiomerne čistoće. Tako se u ovom radu ispitivala mogućnost primjene dva pristupa zelene tehnologije u sintezi (R)-1-feniletanola: (i) primjena lipaze B kao biokatalizatora u hidrolizi (R, S)-1-feniletil-acetata (ii) primjena prirodnih eutektičkih otapalima na bazi kolin-klorida kao medija za provođenje hidrolize (R, S)-1-feniletil-acetata. Praćenjem parametara (η, ee), zaključuje se da je najpovoljnije otapalo za lipazom kataliziranu enantioselektivnu hidrolizu prirodno eutektičko otapalo kolin-klorid:glukoza s 50 % (v/v) vode (ChGlc50). Nadalje, optimirana je reakcija enantioselektivne hidrolize (R, S)-1-feniletilacetata u prirodnom eutektičkom ChGlc50. Optimirana je temperatura reakcije te omjer enzima i supstrata. Istražen je i mehanizam reakcije te je određen utjecaj alternativnih izvora energije (ultrazvuka i mikrovalova) na tijek reakcije.

Abstract (english)

Natural eutectic solvents can be applied in biotransformations for the production of biologically active chiral compounds or chiral syntones of high enantiomeric purity. In this graduate thesis it is explored the possibility of applying two approaches to green technology in synthesis of (R) -1-phenylethanol: (i) the use of lipase B as a hydrolysis (R, S) -1-phenylethyl acetate hydrolysis (ii) applying aprioric eutectic solvents to choline chloride base as a medium for the hydrolysis (R, S) -1-phenylethyl acetate. By monitoring parameters (η, ee) it is concluded that the most preferred solvent for lipase-catalyzed enantioselective hydrolysis is a natural eutectic solvent choline chloride: 50 % (v / v) water glucose (ChGlc50). Furthermore, the enantioselective hydrolysis reaction (R, S)-1-phenylethyl acetate in the natural eutectic ChGlc50 was optimized. The reaction temperature and the enzyme and substrate ratio are optimized. The mechanism of reaction was also studied and the influence of alternative energy sources (ultrasound and microwaves) on the reaction course was determined.
