Abstract

Evolviabilnost i prirodna selekcija omogućavaju bakterijskim stanicama život u nepovoljnim uvjetima rasta. Tijekom produljene stacionarne faze rasta u genomu bakterijskih stanica otkrivene su brojne mutacije i prerasporedi. U ovom radu praćen je rast bakterije Lactobacillus plantarum L73 u bogatoj hranjivoj podlozi (MRS) šaržnim uzgojom te su analizirane stanice koje su preživjele desetodnevni uzgoj. U deset dana starim stanicama L. plantarum utvrđene su promijene u sastavu ukupnih proteina dok je sastav plazmida ostao isti. Iste stanice su značajno osjetljivije na sniženu vrijednost pH od 2.8 i pokazuju povećanu otpornost na antibiotike azitromicin i ceftazidin, a osjetljivije su na ceftriaxon. Nadalje spontani mutanti bakterije L. plantarum L73 s mogućnošću rasta na salicinu ili rifampicinu tijekom petnaestodnevnog uzgoja u siromašnoj podlozi (MML) su pokazali različitu dinamiku rasta.

Abstract (english)

Evolvability and natural selection allowed bacterial cells to live in adverse conditions. During the prolonged stationary growth phase various mutations and rearrangements were detected in the bacterial genome. In this study, the growth of bacteria Lactobacillus plantarum L73 in a rich nutrient medium (MRS) was examined. L. plantarum growth was tested by batch cultivation and then the cells that had survived ten-day cultivation were investigated. Ten day-old cells of L. plantarum were determined to have some changes in protein composition while the plasmids remained the same. Also the same cells showed a higher susceptibility to lower pH (2.8) as well as higher resistance to azitromicine and ceftazidime, but a slight susceptibility to ceftriaxone. Furthermore, the spontaneous mutants of bacterium L. plantarum L73 with the possibility of growing on the salicin or rifampicin in minimal medium (MML) during the fifteen day growth, showed different growth dynamics.
