Abstract

Korištenje fosilnih goriva ima neupitan negativan učinak na okoliš, zdravlje ljudi i promjenu klime. S obzirom da smo suočeni s njihovim skorim iscrpljivanjem, javlja se potreba za alternativnim izvorima energije kao što je lignocelulozna biomasa koja je sveprisutna i jeftina. Kompleksnost strukture ove sirovine otežava njezinu razgradnju do jednostavnih tvari što ograničava njezin u primjenu u industrijskim procesima. Cilj ovog rada bio je pronaći pogodne uvjete za predobradu lignocelulozne sirovine otpadne morske trave Posidonia oceanica te optimirati enzimsku hidrolizu kako bi se dobio visok prinos fermentabilnih šećera za uzgoj oleaginoznih mikroorganizama. Najveća učinkovitost postignuta je predobradom sirovine 0,3 M sumpornom kiselinom (24 sata na 80 °C) nakon koje je slijedila enzimska hidroliza s 15 FPU celulaza/g glukana i enzimskim pripravkom Viscozyme u omjeru 1:1 (mL/mL) u odnosu na dodane celulaze. Na hidrolizatima i filtratu kiselinske predobrade uspješno je proveden uzgoj kvasca Rhodotorula sp. i mikroalge Schizochytrium sp. te su proizvedeni lipidi.

Abstract (english)

The use of fossil fuels has a negative effect on the environment, human health and climate change. Considering their soon exhaustion, there is a need for alternative sources of energy, such as lignocellulosic biomass, which is ubiquitous and cheap. Complex structure of this biomass makes its decomposition to simple compounds difficult and limits use in industrial processes. The goal of this experiment was to find convenient conditions for pretreatment of waste seagrass Posidonia oceanica and optimize enzymatic hydrolysis to get a high yield of fermentable sugars for cultivation of oleaginous microorganisms. The highest efficiency was achieved by pretreatment with 0,3 M sulfuric acid (24h at 80 °C) followed by enzymatic hydrolysis with 15 FPU cellulases/g glucan and enzyme mixture Viscozyme in volume ratio 1:1 (mL/mL). Cultivation of yeast Rhodotorula sp. and microalgae Schizochytrium sp. was successfully carried out in the hydrolysates and acid pretreatment filtrate and lipids were produced.
