Abstract

Drosophila melanogaster pokazuje veliki potencijal u istraživanju nespecifičnog imuniteta s mogućom primjenom kod sisavaca. Cilj studije bio je otkriti epigenetičku kontrolu kroz modifikacije histona H3 (acetilacija i fosforilacija) tijekom Pseudomonas infekcije. Dva klinička soja bakterije su korištena, PA14 wt i PA14 ΔrhlR. Prema in vivo istraživanjima, quorum sensing faktor RhlR smatra se odgovornim za virulentnost Pseudomonas aeruginosa bakterije. MOI 1 uzrokovao je prisutnost od 20% inficiranih stanica u infekciji s oba soja, nakon 1h inkubacije. ChIP analize pokazale su značajnu razliku u acetilaciji histona H3 koji je povezan sa upd3 regulacijom u inficiranim stanicama, što može biti indikator aktivacije JAK/STAT signalnog puta tijekom Pseudomonas infekcije. Za razliku od upd3, eiger nije pokazao razliku u acetilaciji, uspoređujući inficirane i neinficirane stanice.

Abstract (english)

Drosophila melanogaster shows great potential in studying innate immunity mechanisms with possible application in mammalians. The aim of this study was to reveal effect of epigenetic control through histone H3 modifications (acetylation and phosphorylation) during Pseudomonas aeruginosa infection. Two clinical PA14 strains are used, wild-type and ΔrhlR. Quorum sensing factor RhlR has been suggested to have a crucial role in Pseudomonas virulence according to in vivo studies. MOI of 1 caused presence of 20% infected cells in both PA14 wt and PA14 ΔrhlR infection after 1h of incubation. ChIP analysis showed significant differences in histone H3 acetylation involved in upd3 gene regulation in infected cells, which can be indicator of JAK/STAT pathway activation due to Pseudomonas infection. Unlike upd3, eiger did not show different level of acetylation in infected comparing to non-infected cells.
