Abstract

Tehnologija rekombinantne DNA obuhvaća skup metoda koje omogućuju konstrukciju rekombinantnih molekula DNA. Primjena ove tehnologije rezultirala je revolucionarnim otkrićima u biologiji i srodnim znanostima. U ovom radu spomenutim metodama konstruirana su dva nova rekombinantna plazmida, pDUK i pDUK2, ugradnjom fragmenta DNA koji sadrži gen kanMX4 iz plazmida pDMS+ u plazmid pDU. Rezultati restrikcijskih analiza potvrdili su uspješnu konstrukciju oba plazmida. Konstruirani plazmidi sadrže ishodište replikacije za bakteriju Escherichia coli, gene koji omogućuju rezistenciju na antibiotike ampicilin i kanamicin te gen URA3 iz kvasca Dekkera bruxellensis.

Abstract (english)

Recombinant DNA technology includes a group of methods that enable construction of recombinant DNA. The application of this technology has resulted in revolutionary discoveries in biology and related sciences. In this study two new recombinant plasmids were constructed, plasmids pDUK and pDUK2 by inserting DNA fragment that contains gene kanMX4 from plasmid pDMS+ in plasmid pDU. The results of restriction analysis confirmed successful construction of both plasmids. Constructed plasmids contain the origin of replication for bacteria Escherichia coli, genes that provide resistance to antibiotics ampicillin and kanamycin and gene URA3 from yeast Dekkera bruxellensis.
