Abstract

β-glukanaze kataliziraju hidrolizu polisaharida građenih od glukoznih jedinica međusobno povezanih β-glikozidnim vezama. β-glukanaze su pretežno ekstracelularni enzimi i izlučuju se u tekući medij pri uzgoju mikroorganizama. Za industrijsku proizvodnju β-glukanaza najviše se upotrebljavaju sojevi bakterije Bacillus subtilus i plijesni Aspergillus niger, a upotrebljavaju se najviše u pivarstvu te u prehrambenoj i fermentacijskoj industriji. U znanstveno-istraživačkim laboratorijima se koriste za protoplastiranje stanica u svrhu izolacije intracelularnih proteina ili prilikom genetskih manipulacija, za ispitivanje svojstava i građe stanične stijenke te izolaciju proteina stanične stijenke. U ovom radu ispitana je mogućnost proizvodnje ekstracelularne β-glukanaze pomoću Candide utilis iz fakultetske zbirke mikroorganizama. Proizvedeni enzim je djelomično pročišćen i koncentriran te mu je aktivnost uspoređena sa aktivnosti komercijalnih preparata glukanaza. Ustanovljeno je da ovaj preparat glukanaze uspješno razgrađuje laminarin, ali slabije djeluje na glukan u intaktnim staničnim stijenkama kvasca.

Abstract (english)

β-glucanase catalyze the hydrolysis of polysaccharides built from glucose units connected by β-glycosidic bonds. β-glucanases are predominantly extracellular enzymes and are excreted into the liquid medium during cultivation of microorganisms. For industrial production of β-glucanase most frequently used strains are Bacillus subtilus and Aspergillus niger. β-glucanases are most frequently used in the brewing and food industry. Furthermore, glucanases are used for research purposes to remove cell wall of microorganisms in order to isolate intracellular proteins or to improve genetic manipulation of cells, for investigating the properties and structure of the cell wall and for isolation of the cell wall proteins. In this paper we investigated the possibility to produce extracellular β-glucanase by Candida utilis from the faculty collection of microorganisms. The produced enzyme was partially purified and concentrated, and its activity was compared with the activity of commercial glucanase preparations. It was determined that glucanase produced by C.albicans successfully degrades laminarin, but has a weaker effect on glucan in the intact yeast cell wall.
