Abstract

U ovom je radu ispitivan način vezanja Scw10p proteina u staničnu stijenku kvasca Saccharomyces cerevisiae. U stijenku se proteini mogu vezati nekovalentno (Scw proteini) i kovalentno (Ccw proteini), a kovalentno se vežu ili preko GPI-sidra i β-1,6-glukana ili esterskom vezom između karboksilne skupine glutaminske kiseline i hidroksilne grupe glukoze (Pirproteini koji se izoliraju alkalnom hidrolizom). U ovom je radu dokazano da se protein Scw10p u stijenku može vezati kovalentno i nekovalentno, ovisno o posttranslacijskim modifikacijama u koje su vjerojatno uključene japsinske proteaze i proteaza Kex2p.

Abstract (english)

This research was focused on the effect of proteolytic processing of Scw10p on the incorporation of protein in the cell wall of Saccharomyces cerevisiae. Yeast cell wall contains proteins that are noncovalently (Scw-proteins) or covalently (Ccw-proteins) bound to β-1,3- glucan, the latter either through GPI-anchors and β-1,6-glucan, or by alkali labile ester linkages between γ-carboxyl groups of glutamic acid and hydroxyl groups of glucoses (Pir-proteins, extracted by mild alkali). In this research it is proved that the Scw10p protein bound in cell wall covalently or noncovalently, depending on posttranslational modification probably by yapsin proteases and by Kex2p protease.
