Abstract | Pripravci gljiva sastavni su dio tradicionalnih medicinskih praksi posebno u Kini i Japanu. U posljednjih deset godina sustavno raste broj istraživanja koja ispituju biološku aktivnost polifenola, flavonoida i polisaharida izoliranih iz gljiva kojima se pripisuju antioksidativna i prooksidativna, antitumorska te imunostimulirajuća svojstva. Cilj rada bio je ispitati citotoksičnost ekstrakata pojedinačnih gljiva i smjesa ekstrakata te utvrditi imaju li prooksidativno djelovanje na humane stanične linije karcinoma malih stanica pluća H69V, adenokarcinoma pluća SKLU-1, karcinoma pločastih stanica pluća SKMES, adenokarcinoma debelog crijeva CACO2 i astrocitoma CCF-STTG1. Provedena je SDS gel elektroforeza te spektrometrija masa radi utvrđivanja identiteta proteina prisutnih u ekstraktima. Rezultati su pokazali da ekstrakti djeluju citotoksično na ispitivane stanične linije. Njihova niža koncentracija pokazala je prooksidativni, a povećanjem koncentracije prevladava antioksidativni učinak. Analiza sastava proteina pokazala je prisustvo brojnih polipeptidnih fragmenata te pozitivna identifikacija nije bila moguća čime je utvrđeno da proteini nisu izvor biološke aktivnosti istraživanih ekstrakata. |

Parallel abstract (English) | Mushroom preparations are integral components of traditional medical practices especially in China and Japan. In the last ten years the number of studies focusing on bioactivity of mushroom polyphenols, flavonoids and polysaccharides displaying combined prooxidative and antioxidative, antitumour and immunostimulating properties is systematically rising. The aim of this study was to analyse the cytotoxicity of individual mushrooms and extract blends and evaluate their prooxidative effect on lung small cell carcinoma H69V, lung adenocarcinoma SKLU-1, lung squamous cell carcinoma SKMES, colon adenocarcinoma CACO2 and astrocytoma CCF-STTG1 human cell lines. SDS gel electrophoresis and mass spectrometry were conducted to determine the identity of inherent proteins. Results confirmed the cytotoxic effect of mushroom extracts on examined cell lines. Lower concentrations displayed prooxidative, while increased concentrations yielded predominately antioxidative effects. Protein content analysis confirmed high quantities of polypeptide fragments thus preventing positive identification and excluding proteins as the source of bioactivity in tested extracts. |
