Abstract

Sulfonamidi su antibiotici upotrjebljavani često u veterinarskoj praksi, najviše zbog širokog spektra antimikrobnog djelovanja te svoje niske cijene. Više od 10 različitih vrsta ovih sintetičkih komponenti primijenjuju se u veterinarskim ljekovima kod tretmana raznih bakterijskih i protozoalnih infekcija u goveda, svinja i peradi. Sulfonamidi kompetitivno inhibiraju pregradnju p-aminobenzojeve kiseline u dihidropteroat, bakterijama neophodan u sintezi folne kiseline, čime dovode do smanjene aktivnosti u sustavu nukleinskih kiselina. Prisutnost ostataka sulfonamida u hrani je od toksikološke i regulatorne važnosti zbog kancerogenosti nekih spojeva i alergijskih reakcija preosjetljivosti te terapeutske učinkovitosti u ĉovjeka. Za određivanje sulfonamida prisutnih i u najmanjim koncentracijama potrebne su izuzetno osjetljive, kvalitetne metode. Jedna od takvih metoda, ujedno precizna i jeftina za rutinsko izvođenje, jeste imunoenzimska metoda. Primjenjiva je u određivanju širokog spektra sulfonamida sa niskom granicom detekcije. Metoda je zakonskim propisima prihvaćena u Republici Hrvatskoj za analizu ostataka antibiotika u uzorcima životinjskog porijekla i temeljito je opisana u ovom radu.

Abstract (english)

Sulfonamides are antibiotics most commonly used in veterinary practices because of their wide-spectrum antimicrobial activity and inexpensiveness. There are more than 10 diferent sorts of these synthetic compounds rutinely used in veterinary medicines to treat a variety of bacterial and protozoan infections in cattle, swine and poultry. Sulfonamides competitively inhibit conversion of p-amino benzoic acid to dihydropteroate, which bacteria need for folic acid synthesis. This leads to a decreased activity in nucleic acid system. The presence of sulfonamide residues in food is of toxicological and regulatory concern as some of them could be carcinogenic and cause allergic hypersensitivity reactions and therapeutic ineffectiveness in human beings. Quality methods are needed for detection of sulfonamides in the smallest concentrations. One of them which is both accurate and inexpensive is enzyme immunoassay method which can be used in determination of wide range of sulfonamides with low limit of detection. This method is accepted by the legislations of the Republic of Croatia for analysis of antibiotic residues in animals and is thoroughly described in this work.
