Abstract

Cilj ovog istraživanja je bio ispitati funkcionalnu ulogu S-layer proteina (SlpA) probiotičkog soja Lactobacillus helveticus M92 u kompetitivnoj ekskluziji Salmonella enterica subsp. Typhimurium FP1. Prethodna istraživanja upućuju na mogućnost smanjenja infekcije bakterijom S. Typhimurium FP1, kod eksperimentalnih miševa, nakon oralne primjene probiotičkog soja L. helveticus M92, što može biti posljedica kompetitivne ekskluzije ovog soja u gastrointestinalnom traktu (GIT). U ovom radu je ispitan mikrobni antagonizam probiotičkog soja L. helveticus M92 prema S. Typhimurium FP1, te je ustanovljeno da L. helveticus M92 inhibira rast S. Typhimurium FP1 tijekom 24 sata uzgoja. Stoga je ispitana i koagregacija ovog probiotičkog soja sa S. Typhimurium FP1. Provedena su i istraživanja adhezije L. helveticus M92 na Caco-2 epitelne stanice. Ispitana je i mogućnost smanjenja infekcije Caco-2 epitelnih stanica sa S. Typhimurium FP1 nakon primjene probiotičkog soja L. helveticus M92, te uloga S-layer proteina. Preliminarni rezultati pokazali su da primjena probiotičkog soja L. helveticus M92 inhibira invaziju patogene bakterije S. Typhimurium FP1 na Caco-2 epitelne stanice.

Abstract (english)

The objective of this study was to investigate the functional role of the S-layer protein (SlpA) from probiotic strain Lactobacillus helveticus M92 in the competitive exclusion of Salmonella enterica subsp. Typhimurium FP1. The previous researches confirmed that after the oral treatment of experimental mice, infected with S. Typhimurium FP1, with the probiotic strain L. helveticus M92, the pathogen load of Salmonella was lower, as a result of the competitive exclusion of this strain in intestinal tract conferred by probiotic. In this study, microbial antagonism of L. helveticus M92 against S. Typhimurium FP1 was investigated. L. helveticus M92 reduced the growth of S. Typhimurium FP1 during the 24 hours. Therefore, the coaggregation of S. Typhimurium FP1 with L. helveticus M92 was investigated as well. Also, investigation of the adhesion ability of L. helveticus M92 cells on Caco-2 epithelium cells, as well as the possibility to decrease the infection of the Caco-2 cells with S. Typhimurium FP1 after treatment of probiotic strain L. helveticus M92 and the role of S-layer protein in this mechanism were investigated. Preliminary results have shown that application of L. helveticus M92 inhibits in vitro invasion of the S. Typhimurium FP1cells on Caco-2 epihtelium cells.
