Utjecaj inhibitora sinteze stanične stijenke kvasca Saccharomyces cerevisiae na njenu poroznost

Abstract

Stanična stijenka kvasca Saccharomyces cerevisiae je složena struktura, koja stanici daje oblik i čvrstoću te selektivno propušta različite molekule u i iz stanice. Propusnost stijenke ovisi prvenstveno o svojstvima manoproteinskog sloja te o aktivnosti glukanaza i transglikozidaza koje izgrađuju i razgrađuju glukanski sloj stijenke. Nedostatak aktivnosti ili povećana aktivnost ovih enzima u stijenci bi stoga mogla utjecati na njenu poroznost, kao i inhibitori sinteze stijenke i sredstva koja djeluju na interakcije molekula koje ju izgrađuju. U ovom radu je razrađena metoda za ispitivanje poroznosti stijenke praćenjem izlučivanja periplazmatskog enzima kvasca, kisele fosfataze, u medij tijekom uzgoja kvasca ili nakon tretmana stanica reagensima za koje se smatra da utječu na poroznost stijenke. Ustanovljeno je da nedostatak Scw4p proteina u stijenci smanjuje, a suvišak Scw4p povećava propusnost stijenke. Nadalje je utvrđeno da djelovanje inhibitora sinteze stijenke Congo red, Calcofluor white i ditiotreitola (DTT) tijekom uzgoja stanica smanjuje dok tretman stanica Li-acetatom i DTT-om povećava poroznost stanične stijenke.

Abstract (english)

Saccharomyces cerevisiae cell wall has a complex structure which gives the cell its shape and firmness and it selectively penetrates different molecules to and from the cell. Cell wall permeability depends on the features of the manoprotein layer and the activity of the glucanase and transglicosidase which build and disintegrate the glucan layer of the cell wall. Insufficient or overactivity of these enzymes could therefore influence its porosity, and so could the inhibitors of the cell wall synthesis which affect the interactions of the molecules which they build. This thesis discusses the method of cell wall porosity testing through monitoring of extraction of the yeast periplasmatic enzyme, acide phosphatase into the medium during the yeast cultivation or after the treatment of cells with reagents which are considered to affect the cell wall porosity. It has been established that the lack of Scw4p protein in the cell wall reduces and the surplus of Scw4p increases the cell wall permeability. It has also been found that the effect of wall synthesis inhibitors Congo red, Calcofluor white and ditiotritol (DDT) reduces the cell wall porosity while the treatment of cells with Liacetate and DDT increases it.
