Abstract

Cilj rada je ispitati strukturnu nestabilnost mikrosatelitnog markera BAT-26 gena hMSH2. Istraživanje se radilo na uzorku od 20 pacijenata s tumorom zametnih stanica testisa. Lančana reakcija polimeraze (PCR) se koristila kao metoda umnožavanja regije mikrosatelitnog markera BAT-26 hMSH2 gena, a metoda gel elektroforeze se koristila za pregled dobivenih rezultata. Rezultat istraživanja je pojava mikrosatelitne nestabilnosti u samo jednom uzorku, gubitak heterozigotnosti (LOH) nije uočen ni u jednom uzorku. Prema rezultatima moglo bi se zaljučiti da je regija mikrosatelita BAT-26 unutar gena hMSH2 stabilna.

Abstract (english)

The purpose of this work was to examine the structural instability of BAT-26, a microsatellite marker of MSH2 gene. The research involved a sample of 20 patients with testicular germ cell tumor. Polymerase chain reaction (PCR) was used as a method for multipying the regions of microsatellite marker BAT-26 of gene hMSH2, and a method of gel electrophoresis was used to view results. The results showed a phenomenon of microsatellite instability in just one sample and the loss of heterozygosity (LOH) was not seen in any of the samples. According to the results, the microsatellite region BAT-26 in the MSH2 gene, is stable.
