Abstract

Cilj rada bio je praćenje parametara tijekom proizvodnje mupirocina u svrhu praćenja kvalitete proizvoda. Provedeno je praćenje: vode po Karl Fischeru, optičke gustoće na spektrofotometru, amonijačnog dušika na Buchi-ju, pH na pH metru, sadržaj kiseline i lužine standardnom titracijom, određivanje reducirajućih šećera retitracijom, određivanje topljivosti i stranih čestica. Metodom plinske kromatografije potrebno je kvantitativno odrediti sadržaj metil-izobutil ketona (MIBK) i n-heptana u pojedinim frakcijama regeneracije otapala u proizvodnji Mupirocina. Mupirocin je antibiotik koji se proizvodi pomoću mikroorganizma Pseudomonas fluorescens. Regulacija biosinteze mupirocina ovisna je o cijelom nizu parametara koji moraju biti u točno određenim granicama, te u strogo kontroliranim uvjetima.

Abstract (english)

The main goal during the biosynthesis of mupirocine was tracking the parameters in the cause of product quality. Paramethers that have been defined are: water by Karl Fischer, optical density on spectrophotometer, ammonium nitrogen on Buchi, pH on pH-meter, the amount of acid and alkalies by standard titration, defining reduced sugars by retitration, defining solubility and foreign specks. By the method of the gas chromatography is needed to quantitatively determinate the amount of methylisobutyl ketone (MIBK) and n-Heptane in specific fractions of regeneration of the solvent in biosynthesis of mupirocin. Mupirocin is an antibiotic which is produced by microorganism Pseudomonas fluorescens. Regulation of biosynthesis of mupirocine depends on the whole specter of paramethers which have to be in exactly determinated limits and in strictly controlled conditions.
