Određivanje citokina i β-defenzina elisa tehnikom u djece s akutnom respiratornom infekcijom

Abstract

Respiratorni sustav neprestano je izložen brojnim štetnim agensima, mikroorganizmima, česticama i alergenima. U uklanjanju i obrani domaćina od navedenih čimbenika sudjeluju komponente nespecifične i specifične imunosti. Nespecifična imunost uključuje brojne mehanizme, a jedan od njih su antimikrobni peptidi koji posjeduju sposobnost inaktivacije različitih mikroorganizama. Oni mogu biti stalno prisutni ili potaknuti na ekspresiju u epitelnim stanicama (posebice u epitelu dišnih puteva). Važna porodica antimikrobnih peptida su β-defenzini. Epitelne stanice izvor su konstitutivno eksprimiranog β-defenzina-1 dok proupalni citokini potiču ekspresiju β-defenzina-2. U ovom istraživanju, odredili smo povišenu koncentraciju β-defenzina-1 i β- defenzina-2 u uzorcima bronhoalveolarnih ispiraka djece s akutnom respiratornom infekcijom uzrokovanom respiratornim sincicijskim virusom, u odnosu na bakterijama uzrokovanu infekciju. Iako nismo našli statističke razlike u koncentracijama proupalnih citokina TNF-α, IL-1β i IL-6 između djece s virusnom i bakterijskom infekcijom, ustvrdili smo pozitivnu korelaciju ekspresije humanog β-defenzina-1 i TNF-α te humanog β-defenzina-2 i IL-1β.

Abstract (english)

The respiratory tract is continuously exposed to numerous noxious agents, microbial organisms and allergens. Components of innate and specific defense mechanisms participate in elimination and host defense of mentioned factors. The innate host defense include many mechanisms, and one of them are antimicrobial peptides. They are widely expressed and rapidly induced at epithelial surfaces (expecially in respiratory system). Important family of antimicrobial peptides are β-defensins. The epithelial cells are a source of constitutively produced β-defensin-1 whereas proinflammatory cytokines induce expression of β-defensin-2. In this study,we determined increased levels of hBD1 and hBD2 in bronchoalveolar lavate samples from children with the acute virus respiratory infection compared to the bacterial respiratory infection group. Although we have not found statistically different TNF-α, IL-1β and IL-6 levels between children with viral or bacterial respiratory infections, there was a positive correlation between hBD1 and TNF-α as well hBD2 and IL-1β levels in BAL.
