Abstract

Cilj rada bio je validirati enzimatsku metodu određivanja glukoze i fruktoze u vinu SIA-tehnikom. Određeni su parametri: linearnost, ponovljivost mjerenja, ponovljivost pripreme standardne otopine, točnost, limit detekcije i limit kvantifikacije. Dobiveni rezultati su pokazali da je metoda točna, precizna te pogodna za ovu vrstu analize. Nakon validacije su uzorci crnih i bijelih vina analizirani enzimatskom i klasičnom metodom. Dobiveni rezultati su uspoređeni te su pokazali visoku korelaciju navedenih metoda. Pri tom se enzimatska metoda pokazala bržom, jednostavnijom i jeftinijom.

Abstract (english)

The aim of this study was to validate the enzymatic method for determination of glucose and fructose in wine with SIA-technique. Determined parameters were: linearity, repeatability of measurements, repeatability of standard solution preparation, accuracy, limit of detection and limit of quantification. The results obtained showed that the method was accurate, precise and suitable for this type of analysis. After validation, the samples of red and white wines were analyzed by enzymatic and classical method. These results were compared and showed a high correlation between these methods. Besides, enzymatic method showed a faster, simpler and low-cost.
