Abstract

U ovom radu je katakteriziran rekombinantni protein Rny1p/Ccw12p koji je eksprimiran na staničnoj stijenci kvasca Saccharomyces cerevisiae. Stanična stijenka je organela koja stanici kvasca daje mehaničku i osmotsku stabilnost. Proteini mogu biti vezani na staničnu stijenku kovalentno ili nekovalentno. Kovalentna imobilizacija heterolognih proteina na staničnoj stijenci kvasca ima vrlo važnu ulogu u farmaceutskoj, kemijskoj, prehrambenoj industriji i u industriji detergenata. Protein karakteriziran u ovom radu je rekombinantni protein Rny1p/Ccw12p. Nativni Rny1 protein spada u skupinu T2RNaza u kvascu, ali homolozi ovog proteina pronađeni su i u svim drugim organizmima od proteozoa do životinja. U ovom radu ispitana je lokalizacija dobivenog rekombinantnog proteina, njegova aktivnost, stabilnost i određeni su optimalni pH i temperatura za provođenje enzimske reakcije.

Abstract (english)

In this work the recombinant protein Rny1p/Ccw12p which is expressed on the cell wall of yeast Saccharomyces cerevisiae is characterised. The cell wall is the organelle which provide mechanical and osmotic stability of the yeast cell. Proteins can be bound to the cell wall covalently or noncovalently. Covalent immobilization of heterologous proteins on yeast cell wall plays an important role in the pharmaceutical, chemical, food processing industry and industry of detergents. Native Rny1 protein belongs to the group T2RNase in yeast, homologues of this protein were found in all other organisms. This work examined the localization of the recombinant protein, its activity and stability, determineted optimum pH and temperature for the enzymatic reaction.
