Abstract

Peptidi nisu pogodni za korištenje u terapeutske svrhe iz brojnih razloga. U cilju poboljšanja nepovoljnih svojstava njihove se strukture modificiraju, a nastali spojevi nazivaju se peptidomimetici. Oni oponašaju strukturu i biološki učinak peptida te se mogu koristiti kao lijekovi. Autoimune bolesti posljedica su nemogućnosti razlikovanja vlastitih stanica od stranih antigena pri čemu organizam uništava vlastito tkivo. U terapiji autoimunih bolesti koriste se imunomodulatori. Multipla skleroza autioimuna je bolest kod koje se razara mijelin unutar središnjeg živčanog sustava što rezultira nizom simptoma. Mimetici peptidnih epitopa, supresijom razgradnje mijelina, uspješno se koriste u liječenju multiple skleroze. Liječenju karcinoma pristupa se antigen-specifičnom imunoterapijom čiji je cilj omogućiti stanicama imunosustava prepoznavanje tumorskih stanica i selektivno uništavanje.

Abstract (english)

The peptides are not suitable for therapeutic application for various reasons. Their structures are modified to improve the unfavorable properties and the resulting compounds are named peptidomimetics. They mimic the structure and biological effect of the peptides and can be used as medicaments. Autoimmune diseases are the result of inability to distinguish its own cells from foreign antigens in which the body destroys its own tissue. The immunomodulators are employed in the treatment of autoimmune diseases. Multiple sclerosis is an autoimmune disease that destroys myelin within the central nervous system resulting in a set of symptoms. The protein epitope mimetics are successfully applied in treatment of multiple sclerosis owing to their role in the suppression of degradation of myelin. One of the cancer treatment approaches is based on the antigen-specific immunotherapy, which aims to enable the cells of the immune system to recognise and selectively destroy the tumor cells.
