Primjena CCO stanične linije u istraživanjima citotoksičnosti ionskih tekućina

Abstract

Interakcije kemijskih kontaminanata i bioloških sustava najprije se odvijaju na staničnoj razini pri čemu primjena in vitro staničnih kultura može biti vrijedan alat za ranu detekciju kemijskog izlaganja. Posljednjih godina ionske tekućine zaokupljaju pažnju znanstvenika i industrije jer se koriste kao zamjena za hlapiva organska otapala (VOCs). Obzirom na nisku hlapljivost i nezapaljivost, postojane su u okolišu pa je prije šire primjene potrebno odrediti njihovu moguću toksičnost. U ovom radu prikazani su rezultati toksičnih učinaka odabranih ionskih tekućina na proliferaciju stanične linije CCO određeni WST-1 metodom. Na temelju izračunatih EC50 vrijednosti redoslijed toksičnosti odabranih imidazolijevih ionskih tekućina bio je: [C10MIM][Tf2N] > [C7MIM][Tf2N] > [C3- 3MIM][Tf2N] > [C3-2MIM][Tf2N] > [C4MIM][Tf2N] > [C4MIM][PF6] > [C4MIM][BF4]. Toksični učinci ispitanih ionskih tekućina na stanicama CCO bili su mnogo izraženiji od učinaka konvencionalnih organskih otapala. Morfološke promjene inducirane djelovanjem ionskih tekućina na stanice CCO pokazale su promjene u monosloju i strukturi stanica, a zapažena citotoksičnost uglavnom je bila posljedica nekroze.

Abstract (english)

Interactions of chemical contaminants and biological systems at first take place at the cellular level where the application of in vitro cell cultures can be a valuable tool for early detection of chemical exposure. Recently, ionic liquids occupy the attention of scientists and industry for usage as the replacements for volatile organic compounds (VOCs). Considering their low volatility and inflammability they are persistant in the environment. Therefore it is necessary to determine their possible toxicity before their wider application. This paper presents the results of toxic effects of selected ionic liquids on the proliferation of fish cell line CCO determined by the WST-1 method. Based on the calculated EC50 values the order of toxicity of selected imidazolium ionic liquids was: [C10MIM][Tf2N] > [C7MIM][Tf2N] > [C3- 3MIM][Tf2N] > [C3-2MIM][Tf2N] > [C4MIM][Tf2N] > [C4MIM][PF6] > [C4MIM][BF4]. Toxic effects of ionic liquids tested on cells CCO were much more expressed than the effects of conventional organic solvents. Morphological changes induced by the action od ionic liquids on cells CCO have shown changes on the structure of monolayers of cells while the noticed cytotoxicity was largely the result of necrosis.
