Utjecaj ionskih tekućina na proliferaciju i morfološke promjene CCO stanica

Abstract

Najvažnije svojstvo ionskih tekućina je niska hlapivost zbog čega su predložene kao zelena alternativa tradicionalnim organskim otapalima u cilju smanjivanja njihove difuzije u atmosferu. Za sada postoji relativno ograničen broj istraživanja o toksičnim učincima ionskih tekućina koji su neophodni za sveobuhvatnu procjenu rizika njihova korištenja. In vitro sustavi sa staničnim linijama riba važni su u istraživanjima toksičnog učinka različitih spojeva iz okoliša. U ovom radu prikazani su rezultati citotoksičnosti odabranih ionskih tekućina različite duljine alkilnog lanca na proliferaciju stanične linije CCO pomoću MTT metode. Na temelju izračunatih EC50 vrijednosti redoslijed toksičnosti ionskih tekućina bio je sljedeći: [C7M2IM][Tf2N]>[BzMIM][Tf2N]>[C7MIM][Tf2N]>[C5MIM][Tf2N]>[ApiMIM][Tf2N]> [C4MIM[Tf2N]. Toksičnost ispitanih ionskih tekućina u stanicama CCO bila je izraženija od toksičnosti diklormetana, etanola, DMSO i toluena kao klasičnih organskih otapala. Morfološke promjene pod djelovanjem ionskih tekućina očituju se izmijenjenom monosloju i strukturi stanica, pojavi apoptoze i nekroze, a stanična smrt je uglavnom bila posljedica nekroze.

Abstract (english)

The most important property of ionic liquids is low volatility because of this property were proposed as a green alternative to traditional organic solvents in order to minimize their diffusion in the atmosphere. For now, there is a relatively limited number of studies on the toxic effects of ionic liquids that are essential for a comprehensive assessment of the risks of their use. In vitro systems with cell lines of fish are important in studies of toxicity of different compounds in the environment. This thesis presents the results of cytotoxicity of selected ionic liquids with different lengths Alkyl Chain on the proliferation of CCO cell line using MTT method. Based on the calculated EC50 values of the order of toxicity of ionic liquids was as follows: [C7M2IM] [Tf2N]> [BzMIM] [Tf2N]> [C7MIM] [Tf2N]> [C5MIM] [Tf2N]> [ApiMIM] [Tf2N]> [C4MIM [Tf2N]. Toxicity of ionic liquids tested in CCO cells was stronger than the toxicity of dichloromethane, ethanol, DMSO and toluene as a classical organic solvents. Morphological changes under the influence ionic liquids manifest altered monolayers and cell structure, apoptosis and necrosis occurs, and cell death was mainly due to necrosis.
