Abstract
Endonukleaza I-SceI pripada skupini „homing“ endonukleaza, prepoznaje i cijepa asimetričnu sekvenciju DNA duljine od 18 pb. Ovaj enzim je visoko specifičan, zbog čega se često koristi u genetičkom inženjerstvu i genskoj terapiji. U ovom radu plazmidna DNA izolirana je iz soja kvasca Saccharomyces cerevisiae koji je prethodno transformiran plazmidom koji kodira za enzim I-SceI. Rezultati restrikcijske analize potvrdili su prisutnost plazmida pYES-SCE.
Abstract (english)
Endonuclease I-SceI belongs to the group of „homing“ endonucleases, that recognize and cleave asymmetric 18 pb sequence. As this enzyme has a high specificity, it has been used in genetic engineering and gene therapy. In this study plasmid DNA was isolated from the strain of yeast Saccharomyces cerevisiae, previously transformed by plasmid encoding I-SceI enzyme. The restriction analysis of the plasmid confirmed the presence of pYES-SCE plasmid.
