Abstract

Bakterije u svojem prirodnom okruženju pri izlaganju promjenjivim okolišnim uvjetima podliježu neprestanim promjenama genetičkog materijala. Promjene mogu nastati spontano, ili mogu biti uzrokovane adaptivnom mutagenezom, hipermutacijama ili horizontalnim prijenosom gena. Mutacije su osnovni pokretač evolucije i ključne su za preživljavanje bakterija u stresnim uvjetima. U ovom radu pratio se rast spontanih mutanata bakterija mliječne kiseline u produljenoj stacionarnoj fazi u usporedbi s bakterijama divljeg tipa. Stabilnost genetičkog materijala mutanata ispitivana je metodom gel elektroforeze u pulsirajućem polju. Dobiveni rezultati pokazuju da su mutanti stabilniji i prilagodljiviji naspram stanica divljeg tipa tijekom uzgoja u produljenoj stacionarnoj fazi rasta dok analiza genoma bakterija nije pokazala značajne razlike između mutanata i stanica divljeg tipa.

Abstract (english)

While growing in their natural enviroment, bacteria are exposed to commutable enviromental conditions causing their genetic material to change. Such alterations can be the consequence of spontaneous mutatios or caused by adaptive mutagenesis, hypermutagenesis or by horizontal gene transfer. Mutations are the basic promoter of evolution and are essential for the survival of bacteria growing in a stressful enviroment. The aim of this study was to compare the growth of spontaneous mutants of lactic acid bacteria to the growth of their respective wild type strains. The stability of the mutants' genetic material was analysed by the pulsed field gel electrophoresis method. The results show that the mutants are more stable and adjustable in comparison to the wild type strain during their growth in the prolonged stationary phase while the gel analisys didn't show any significant differences between the mutants' genome and the one belonging to the wild type bacteria.
