Abstract

Svrha rada bila je ispitati prisutnost gubitka heterozigotnosti (LOH) i mikrosatelitne nestabilnosti (MSI) u uzorcima tkiva od dvadeset pacijenata, od kojih je deset imalo seminomne, dok je ostalih deset imalo neseminomne tumore. Pritom smo koristili tehniku umnažanja DNA PCR-om intragenskog mikrosatelitnog markera D17S855 u genu BRCA1, te gel elektroforezu za iščitavanje rezultata. Na svim uzorcima pacijenata primjećena je informativna heterozigotnost, a kod tri od dvadeset uzoraka prisutna je mikrosatelitna nestabilnost u obliku skraćenog alela. Uočena MSI može biti dio ukupne genomske nestabilnosti tih tumora, iako su dobiveni samo preliminarni rezultati.

Abstract (english)

The aim of the study was to examine the presence of loss of heterozygosity (LOH) and microsatellite instabillity (MSI) in tissue samples from twenty patients, of whom ten had seminomal, while the other ten had nonseminomal tumors. Thereby, we used the PCR technique to amplify intragenic mikrosatellite marker D17S855 within BRCA1 gene and gel electrophoresis in order to visualize results and to draw conclusion. Informative heterozigosity was observed in all patient samples and in three of twenty samples, microsatellite instabillity is present in the form of shortened allel. Results are preliminary but observed MSI may be integral part of inherent genomic instability in those tumors.
