Abstract

HEK (ljudska embrionalna bubrežna) stanična kultura se koristi za proizvodnju biološki aktivnih tvari za medicinske i dijagnostičke potrebe te je poznavanje uvjeta uzgoja ove stanicne linije neophodno za povećanje učinkovitosti proizvodnih postupaka. Cilj ovog rada je istražiti kako glukoza i glutamin, kao dva najznačajnija izvora energije, djeluju na rast HEK stanica i ustanoviti koji je od tih sastojaka značajniji za održavanje kulture stanica. Stanice smo uzgajali u četiri različita medija, sa ili bez glukoze i glutamina. Istraživanje je pokazalo da su za uspješan rast i razvoj stanica neophodni i glukoza i glutamin te da su podjednako važne komponente hranjivog medija.

Abstract (english)

HEK (Human Embryonic Kidney) cell culture is used for the production of biologically active substances for medical and diagnostic purposes. The aim of this study was to examine the effect of glucose and glutamine on the growth of HEK cells. To determine which of these two nutrients had greater impact on cell growth, cells were cultivated in four different culture media, with or without glucose and glutamine added. The methods we used were cell cultivation in controlled conditions and the cell number determination over a period of eight days. Analysis of culture media was performed. The results showed that glucose and glutamine are equally important components of a medium for growth and development of the cells.
