Abstract

Listeria monocytogenes je široko rasprostranjena u okolišu zahvaljujući svojim sposobnostima da podnosi i raste u različitim vanjskim uvjetima. Ipak, trenutna saznanja o ekologiji ove bakterijske vrste, posebno o načinima preživljavanja i prijenosa u prirodnom okruženju, još su uvijek djelomična i nepotpuna. Cilj ovog rada bio je istražiti interakciju između protozoe Acanthamoeba castellanii i L. monocytogenes u in vitro uvjetima. Rezultati su pokazali da L. monocytogenes ne preživljava u ovoj vrsti ameba, obzirom da već nakon 5-6h nismo uspjeli izolirati unutarstanične bakterije iz ko-kulture. Ipak, vanstanične listerije imale su koristi u ko-kulturi s amebom, jer im se povećao broj u odnosu na kontrolu. Zaključno, L. monocytogenes ne može preživjeti unutar A. castellanii u ispitivanim laboratorijskim uvjetima. Sudeći prema dobivenim rezultatima, A. castellanii ne predstavlja prirodni okolišni rezervoar za L. monocytogenes, ali tijekom ko-kultivacije, svojim prisustvom utječe na razmnožavanje i doprinosi preživljavanju vanstanične listerije.

Abstract (english)

Listeria monocytogenes is found ubiquitously in the environment and is capable to grow and survive in a wide range of environmental conditions. However, the current knowledge of its ecology and in particular, the mode of its environmental survival and transmission remains limited. The aim of this study was to investigate the interaction between the protozoan Acanthamoeba castellanii and Listeria monocytogenes. The results indicate that L. monocytogenes does not reside within A. castellanii, as we were unable to recover any intracellular bacteria after approximately 5 hours of co-cultivation. However, extracellular bacteria benefit from co-cultivation with amoeba showing increased growth and multiplication. In conclusion, Listeria monocytogenes is not able to survive inside Acanthamoeba castellanii, but is quickly degraded under the laboratory conditions tested. A. castellanii is not able to act as environmental reservoir for L. monocytogenes but may provide a survival advantage for extracellular listeria during co-cultivation of the two organisms.
