Abstract

U posljednje se vrijeme, u javnosti, pojavilo veliko zanimanje za oblikovanje novih lijekova. Biološki aktivne supstancije za njihovo se oblikovanje sve više traže u genomima i metagenomima sekvenciranih mikroorganizama. Razvijen je čitav niz javno dostupnih baza podataka i bioinformatiĉkih alata za njihovu analizu. To se posebno odnosi na bioinformatiĉke alate koji analiziraju genske nakupine, čiji produkti sudjeluju u sintezi sekundarnih metabolita u uvjetima in silico. U ovome je radu sistematizirano znanje o dosada poznatim i detaljno opisanim sekundarnim metabolitima, neribosomalno sintetiziranih peptida i siderofora, radi njihove bolje klasifikacije. Detaljnija klasifikacija omogućila je izradu vlastitih profila proteina za navedene skupine sekundarnih metabolita kojima je unaprijeđen bioinformatički alat ClustScan. Ti su profili testirani na novosekvencioniranom i dosada neopisanom genomu vrste Streptomyces scabies. Na temelju sličnosti genoma vrste S. scabies i dosad anotiranih genoma S. coelicolor i S. avermitilis pretpostavlja se da će anotiranje novog genoma dovesti do pronalaska novih genskih nakupina odgovornih za biosintezu neribosomalno sintetiziranih peptida i siderofora.

Abstract (english)

During the last several years there has been an increased public interest in the design of novel drugs. Majority of biologically active substances for their design have been largely looked in sequenced microbial genomes and metagenomes. For that reason a number of publically available databases and bioinformatics tolls were developed to analyse gene clusters whose products are involved in the biosynthesis of secondary metabolites in silico. In this work the knowledge of already known secondary metabolites, nonribosomally synthesized peptides and siderophores, was systematized for their better classification. More detailed classification allowed the design of the propriety protein profiles for the gene cluster proteins involved and subsequently used for the improvement of the ClustScan program package. These profiles have been tested on the newly sequenced genome of Streptomyces scabies. Based on the similarity of the S. scabies genome to the already annotated genomes of S. coelicolor and S. avermitilis it was assumed that the annotation of the new genome will lead to the discovery of novel gene clusters responsible for the production of yet unrecognised nonribosomally synthesized peptides and siderophores.
