Abstract

Protein RecA je esencijalni protein homologne rekombinacije bakterije E. coli. Stanice mutirane u genu recA imaju smanjenu vijabilnost. U ovom radu je proučena stabilnost genoma nakon γ-zračenja u stanicama divljeg tipa, te u mutantima bakterije E. coli. Istraživanja su pokazala da mutanti (recA ΔrecBC i recAΔ recBCΔ sbcB) preživljavaju manje doze zračenja od divljeg tipa bakterije te da dolazi do rearanžmana samo u stanicama divljeg tipa. Za analizu genoma koristila se gel elektroforeza u promjenjivom električnom polju (metoda PFGE).

Abstract (english)

RecA protein is an essential protein of the homologous recombination of the E. coli bacterium. Cells mutated in the recA gene have a lowered viability. In this work, the stability of the genome after γ- radiation in wild type cells was studied as well as mutants of the bacterium E. coli. Studies have shown that mutants (recA ΔrecBC i recAΔ recBCΔ sbcB) survive smaller doses of radiation than the wild type bacteria and genome rearrangements have been detected only in the wild type cells. Pulsed-field gel electrophoresis (PFGE) was used for genome analysis.
