Abstract

Metodom ELISA testa i tekućinskom kromatografijom visoke djelotvornosti sa spektrometrom masa (LC-MS/MS) analiziran je uzorak tkiva obogaćen kloramfenikolom i na temelju rezultata usporedene su te dvije analitičke metode. Određeni su parametri iskorištenja, ponovljivosti i linearnosti te drugi parametri koji utječu na učinkovitost određivanja kloramfenikola. Analiziranjem uzoraka rezultati su pokazali veću preciznost i ponovljivost LC-MS/MS metode na svim obogaćenjima uzoraka te iskorištenje i vrlo dobru linearnost. Zbog toga se i ELISA koristi kao orijentacijska metoda, dok LCMS/ MS služi kao potvrdna metoda u slučaju pozitivnog testa uzoraka na prisutnost kloramfenikola.

Abstract (english)

A tissue sample enriched with chloramphenycol was analyzed by using ELISA test and High Performance Liquid Chromatography with Mass Spectrometry (LC-MS/MS) and the results were compared between the two analytical methods. The parameters efficiency, repeatability and linearity and other parameters that affect the efficiency of chloramphenicol analysis were determined. The results obtained by LC-MS/MS method showed greater accuracy and repeatability at all enrichment of samples, as well as recovery and very good linearity. Therefore, the ELISA method is used as orientation method, while LC-MS/MS is used as a confirmatory method in the case of a positive test of chloramphenicol in samples.
