Abstract | Dosadašnja istraživanja na vrsti Astragalus membranaceus pokazala su da se ekstrakt astragalusa do sada nije primjenjivao kao regulator imunološke reakcije, već kao antioksidans i imunostimulator. U istraživanju su korišteni CBA miševi koji su primali 3 različite doze lijeka, koje su im unošene u organizam per os. Cilj istraživanja bio je utvrditi da li je lijek Lectranal® toksičan kod oralne upotrebe te ispitati njegovu učinkovitost u sprječavanju razvoja inflamatornih i alergijskih reakcija putem aktivacije gena koji reguliraju njihov tijek. Tokom akutnog ispitivanja u trajanju od 30 dana praćene su: fenotipske promjene, promjene u ponašanju i preživljavanju, promjene u tjelesnoj težini, promjene biokemijskih parametara te kliničkih kemijskih parametara u urinu, dok je RNA izolirana iz slezene podvrgnuta chip metodi prevođenja u cDNA, te kao takva real- time PCR-u. Rezultati pokazuju da nema razlike između tretiranih i kontrolnih životinja koje bi upućivale na toksični učinak Lectranal®-a, te da njegova primjena aktivira gensku regulaciju inflamatornih procesa uzrokovanih alergenima, što dokazuje da je pripravak učinkovit u sprječavanju razvoja inflamatornih i alergijskih reakcija. |

Parallel abstract (English) | Previous researches had shown that the extract of Astragalus membranaceus hasn't been used as a regulator of immune reactions, but implemented as an antioxidant and immunostimulator. In our research we used CBA mice that received 3 different doses of the medicine Lectranal®, that were entered per os. The aim was to determine if the medicine is toxic in oral use, and to examine its effectivnes in preventing the development of inflammatory and allergic reactions by activation of genes that regulate their flow. During research in the acute period of 30 days there were followed: phenotype changes, changes of behavior and survival, changes of body weight, changes of biochemical parameters and clinical-chemical parameters in urin, while the isolated RNA from the spleen was subjected to the chip method of translating to cDNA, and as such to real-time PCR method. The results show that there is no difference between control and treated animals, that would be indicative to any toxic effect of Lectranal®, and that its application acitvates gene controlling inflammatory process caused by allergens, indicating that the medicine is effective in preventing the development of inflammatory and allergic reactions. |
