Abstract

Usporedene su dvije u literaturi opisane metode MeOH i EtOH za ekstrakciju smjese aristolohičnih kiselina AA I i AA II iz lista, stabljike, korijena, ljuske i sjemenki ploda biljke A. clematitis sakupljenih u različitim djelovima Hrvatske. Kvalitativna analiza izolirane smjese AA I i AA II provedena je visokoučinkovitom tekućinskom kromatografijom obrnutih faza (RP-HPLC). EtOH metoda pokazala se efikasnijom u odnosu na MeOH metodu dajući bolja iskorištenja u sveukupnom ekstraktu kao i na višem udjelu smjese AA I i AA II u sirovom ekstraktu. LC-MS-MS metodom određene su točne molekulske mase za AA I (retencijsko vrijeme 2,13 min) te je nađeno m/z [M+H+]+ = 342,0650, a za AA II (retencijsko vrijeme 1,72 min) nađeno je m/z [M+H+]+ = 312,0527.

Abstract (english)

Two methods reported in literature for extraction of aristolochic acid mixture (AA I and AA II) were compared. Aristolochic acids were extracted with methanol or ethanol (70 %) from the leaf, stem, root, shell and seed grain yield of the plant A. clematitis collected in different regions in Croatia. Qualytative reverse phase high performance liquid chromatography (RP-HPLC) analysis of extracted mixture of AA I and AA II was performed. Ethanol extraction was more efficient yielding higher percentage of entire extract and, as well as, higher share of mixture AA I and AA II in the raw extracts. Applying LC-MS-MS method for AA I (retention time 2,13 min) m/z [M+H+] + = 342.0650 and for AA II (retention time 1,72 min) m/z [M+H+] + = 312.0527 was found.
