Abstract | Likopen , pigment je koji rajčici daje crvenu boju. Uobičajeno određivanje koncentracije likopena ostvaruje se klasičnim metodama poput HPLC-a i spektrofotometrije. Iako su metode apsolutne, zahtjevaju skupu i dugotrajnu ekstrakciju pigmenta prije same analize. U ovome radu, predložen je nov pristup određivanju koncentracije likopena. To su Fotoakusticna spektroskopija(FA) sa argon laserom, Fotoakustična spektroskopija(FA) sa Xe lampom i Rezonantna Ramanova spektroskopija. Osnovna ideja jest istraživanje moguće korelacije između koncentracije likopena i veličine signala mjerenih sa gore navedenim metodama. Različiti proizvodi bazirani na rajčicama su korišteni kao eksperimentalni uzorci. Rezultati FA i Raman mjerenja stavljeni su u odnos sa kolorimetrijskim indeksima L*, a*, b*. Odlična korelacija postignuta je između FA signala (Xe lampom ili laserom) i koncentracije likopena izmjerenom putem HPLC-a. Isto vrijedi za rezultate postignute Ramanovom tehnikom. Nažalost korelacija izmedu L*, a*, b* indeksa i koncentracije likopena nije tako dobra. U ovom trenutku razlozi za opaženu diskrepanciju nisu poznati. |

Parallel abstract (English) | Lycopene, the red pigment naturally present in tomatoes. Concentration of lycopene is usually determined by the absolute methods such as HPLC and spectrophotometry, but the extraction procedure of lycopene is a time consuming, destructive and costly. In this study a new approach to measurement of lycopene is presented. The newly proposed methods used here are the Photoacoustic spectroscopy(PA) with argon laser, Photoacoustic spectroscopy (PA) with Xe lamp and Resonance Raman spectroscopy. Various tomato-based products were used as test samples; all of them contain lycopene as a dominant colorant. This research study shows a good correlation between the spectroscopic techniques and lycopene content. Unfortunately the correlation between the colorimetric indices (a*, b* and L*) and the concentration of lycopene was rather poor. |
