Abstract

Endiinski antitumorski antibiotici već 30 godina bude interes kemičara zbog sposobnosti ciklizacije u vrlo reaktivan 1,4-didehidrobenzenski biradikal, koji posjeduje sposobnost cijepanja dvostruke uzvojnice DNA. Primjećeno je da prisustvo prijelaznih metala utječe na ciklizaciju sintetskih endiinskih sustava. U svrhu dobivanja analoga endiina pogodnih za kompleksiranja metalnih iona, priređeni su i karakterizirani fenilalaninski i alaninski simetrični dipeptidni mimetici premošteni cis-1,8-diaminookt-4-en-2,6-diinskim mostom. Dobiveni endiinski derivati, N1,N8-di(L-fenilalanil)okt-4(Z)-en-2,6-diin (11) i N1,N8-di(L-alanil)okt-4(Z)-en-2,6-diin (12) karakterizirani su i UV/VIS spektroskopijom te je provedena spektrofotometrijska titracija s otopinom Cu2+- iona. Na temelju dobivenih rezultata utvrđeno je nastajanje kompleksa stehiometrije ligand:Cu2+ = 2:1.

Abstract (english)

Enediyne anticancer antibiotics have intrigued chemits for over 30 years because of their abilliy to cyclize to a highly reactive 1,4-didehydrobenzenoid diradical, highly reactive structure capable of the DNA double helix breakage. Transition metals have been shown to influence cyclization of synthetic enediyne compounds. Within the framework of this graduated paper, cis-1,8-diaminooct-4-en-2,6-diyne bridged alanine and phenylalanine symmetrical dipeptide mimetics were prepared and characterized. Spectrophotometric titration of N1,N8-di(L-phenylalanyl)oct-4(Z)-en-2,6-diyne (11), N1N8-di(L-alanyl)oct-4(Z)-en-2,6-diyne (12) with the Cu2+ solution showed formation of complex with ligand : Cu2+ stoichiometry 2:1.
