Utjecaj probiotika i prebiotika na crijevnu mikrofloru i imunološki odgovor u pokusnih miševa

Abstract

U sklopu probiotičkog koncepta, u prethodnim istraživanjima, definirana su probiotička svojstva tri soja bakterija mliječne kiseline: Lactobacillus acidophilus M92, Lactobacillus plantarum L4 i Enterococcus faecium L3. U ovom radu istražene su mogućnosti primjene ovih probiotičkih i različitih prebiotičkih supstrata et njihov utjecaj na crijevnu mikrofloru i imunološki odgovor u pokusnih Swiss Albino miševa. In vivo pokusi na miševima su pokazali da sva tri probiotička soja L. acidophilus M92, L. plantarum L4 i E. faecium L3 preživljavaju uvjete u probavnom sustavu miševa, adheziraju tanko i debelo crijevo, te imaju pozitivan učinak na crijevnu mikrofloru miševa smanjivanjem broja enterobakterija i sulfito-reducrajućih klostrodija. Adhezija ispitivanih probiotičkih sojeva u tankom i debelom crijevu pokusnih miševa potvrđena je induciranjem rezistencije ovih sojeva na rifampicin te identifikacijom izoliranih bakterijskih sojeva iz crijeva miševa RAPD-analizom. Paralelno su provedena istraživanja imunomodulacijskog učinka ispitivanih probiotičkih sojeva i prebiotičkih supstrata na imunološki sustav miševa ELISA metodom. Probiotički sojevi Lactobacillus rhamnosus LGG i Enterococcus faecium L3 su uspješno primjenjene za proizvodnju probiotičkih napitaka od permeata dobivenog ultrafiltracijom mlijeka.

Abstract (english)

In previous researches, in the context of probiotic concept, three strains of lactic acid bacteria with probiotic properties have been defined: Lactobacillus acidophilus M92, Lactobacillus plantarum L4, and Enterococcus faecium L3. In this research, there have been explored many possibilities of appliances to these probiotic strains and various prebiotic substrates and their influence on intestinal microflora and immune response in experimental Swiss Albino mice. In vivo experiments have shown that all three probiotic strains, L. acidophilus M92, L. plantarum L4, and E. faecium L3, survive the gastrointestinal conditions, adhered to small and large intestine of mice and have positive affect on intestinal microflora of mice by reducing the number of enterobacteria and sulphite reducing clostridia. Adhesive function of tested probiotic strains in the small and large intestine of experimental mice has been confirmed by inducing the resistance of these strains on rifampicin, and by identification of isolated bacterial strains out of mice intestine by RAPD-analysis. Parallel with these researches, there have been conducted immunomodulatory effect researches on tested probiotic strains and prebiotic substrates on immunity system of mice by ELISA method. Probiotic strains Lactobacillus rhamnosus LGG and Enterococcus faecium L3 were successfully applied in production of fermented probotic beverage of permeate obtained by ultrafiltrtion of cow´s milk.
