Abstract

Provedena je analiza glukozinolata (GSL) u devet uzoraka sortnog sjemena uljane repice tekućinskom kromatografijom visoke djelotvornosti (HPLC), a UV/VIS spektroskopskim metodama su određeni ukupni polifenolni spojevi i njihova antioksidacijska sposobnost u dvije standardne sorte sjemena uljane repice. Za određivanje glukozinolata upotrijebljena je međunarodna standardna metoda (ISO 1992, 1995) koja uključuje ekstrakciju metanolom, enzimsku desulfataciju i tekućinsku kromatografiju obrnutih faza (RP-LC) nastalih desulfoglukozinolata (DSGSL). Desulfoglukozinolati eluirani s kolone identificirani su prema redoslijedu eluiranja s kolone i analizom UV-spektara, a kvantifikacija je provedena metodom unutarnjeg standarda. Udio ukupnih glukozinolata u istraženim sortama je između 8,31 i 15,37 µmol g-1 sjemena što je u skladu sa standardima Europske unije. Iz odmašćenih standardnih sorti uljane repice pripravljeni su ekstrakti u 70 %-tnom metanolu, koji su poslužili kao uzorci za određivanje antioksidacijske sposobnosti DPPH-metodom. Veću antioksidacijsku aktivnost pokazuju uzorci s većom koncentracijom polifenola.

Abstract (english)

The analysis of rapeseed glucosinolates in nine samples of rapeseed cultivars was done by High-Performance Liquid Chromatography (HPLC), while the presence of polyphenols and their antioxidative activity in two standard rapeseed cultivars was determine by UV/VIS spectroscopy. The method used was Standard method for determination of rapeseed glucosinolates (ISO 1992, 1995). The method includes extraction with methanol, enzymatic desulphation and Reversed-Phase Liquid Chromatography (RP-LC) of desulphoglucosinolates (DSGSL). The identification of desulphoglucosinolates eluted was made according to the elution order and by UV-spectra of the chromatographic peaks, while the quantitative analysis was done by use of the internal standard. The content of total glucosinolates in the cultivars investigated ranged between 8.31 and 15.37 µmol g-1 rapeseed. The extracts of defatted rapeseed samples were prepared in 70 % methanol and used for the determination of total polyphenols by the Folin-Ciocalteau reagent and the estimation of antioxidative activity by DPPH-method. The greatest antioxidative activity was exhibited by the samples with the highest concentration of polyphenols.
