Abstract

Primjenom mikronukleus testa u uvjetima in vitro na ljudskim limfocitima istraženi su toksični učinci triptofola u koncentracijama 0,25mM; 0,50 mM; 1,00 mM i 2,00 mM. Utvrđeno je da triptofol izaziva pojavu mikronukleusa, nuklearnih pupova i nukleoplazmatskih mostova te indukciju programirane stanične smrti ili apoptoze. Navedeni učinci bili su ovisni o koncentraciji i najslabije izraženi u uzorku tretiranom najnižom koncentracijom triptofola (0,25 mM). Dobiveni rezultati ukazuju da je glavnina uočenih oštećenja potaknuta aneugenim učinkom triptofola na limfocite. Tretman triptofolom značajno remeti stanične membrane, uzrokujući smanjenje volumena stanice. Triptofol usporava stanični ciklus u uvjetima in vitro, što potvrđuju vrijednosti indeksa diobe jezgara (NDI), koje se smanjuju usporedno s porastom primijenjene koncentracije. Provedeno istraživanje rasvjetljava dio citotoksičnih učinaka triptofola na ljudske stanice, otvarajući prostor za istraživanja čitavog niza potencijalnih učinaka koji mogu doći do izražaja u organizmu nositelju djelovanjem njegovih metabolita.

Abstract (english)

Toxicity of tryptophol (0.25mM; 0.50 mM; 1.00 mM and 2.00 mM) on human peripheral blood lymphocytes was investigated in vitro using micronucleus assay. Results indicate that tryptophol caused induction of micronuclei, nuclear buds and nucleoplasmic bridges as well as the apoptosis induction. Observed effects were concentration-dependent and their frequencies were the lowest in the sample treated with 0,25 mM of tryptophol. Results obtained indicate that the majority of damages in tryptophol-treated lymphocytes were induced by aneugenic mechanisms. In concentrations tested tryptophol affected cell membranes, which in turn led to the marked decrease of the cell volume. Tryptophol retarded in vitro cell proliferation, and the values of nuclear division indices (NDI), also decreased in parallel with the increase of its concentration. Present study elucidates only a part of tryptophol cytotoxicity to human cells, and leaves open area for further investigations of potential effects in host organism mediated by its secondary metabolites.
