Abstract

Regulacijski T-limfociti čine subpopulaciju T-limfocita vrlo važnih za održavanje periferne imunotolerancije. Njihovu ulogu možemo primjetiti u nizu kliničkih stanja od alergija i autoimunosnih bolesti gdje je opisan smanjeni broj ili funkcionalna sposobnosti ovih stanica, do infekcija pojedinim patogenima i raznih tumorskih oboljenja gdje je situacija suprotna. Dvije su osnovne populacije regulacijskih T-limfocita – prirodni regulacijski T-limfociti koji se do svoje potpune zrelosti razvijaju u timusu, te inducirani regulacijski T-limfociti koji nastaju na periferiji nakon kontakta s antigenom. Istraživanja vezana uz fenotip i mehanizme djelovanja regulacijskih T-limfocita (Treg) od velikog su značaja zbog potencijalnog novog pristupa liječenju čitavog niza kliničkih stanja. Cilj istraživanja u ovom radu bio je odrediti u kojem rasponu se kreću vrijednosti za Treg u zdravih mladih osoba, postoje li razlike u broju Treg između muškaraca i žena, te da li se dobivaju drugačiji rezultati ako se istraživanja provode na svježim stanicama krvi u odnosu na one koje su bile smrznute mjesec dana.

Abstract (english)

Regulatory T cells are subset of T cells that are very important in maintaining of peripheral tolerance. These cells have important role in various clinical conditions such as allergy and autoimmune disorders in which lower number or decreased functional capability of regulatory T cells was described, or different kinds of tumors or infections caused by pathogens where the situation is quite the opposite. There are two main populations of regulatory T cells – natural regulatory T cells (Treg), which fully develop in thymus, and inducible Treg, which develop in the periphery after contact with the antigen. The aim of this study was to determine the number of Treg in healthy, young adults and to investigate the possible influence of gender on the numbers of Treg. We also compared whether cell freezing in liquid nitrogen for one month influenced the frequency of these cells in comparison to fresh samples.
