Abstract

Sulfonamidi su stabilni kemoterapeutici širokog spektra djelovanja koji se u pčelarstvu koriste za prevenciju i liječenje bolesti pčela. Njihova prisutnost u medu u koncentracijama većim od dozvoljenih predstavlja potencijalnu opasnost za ljudsko zdravlje. U Hrvatskoj je dozvoljena koncentracija do 100 µg/kg. Svrha ovog rada bila je optimiranje metode za određivanje sulfonamida u medu. Uzorci su pročišćeni ekstrakcijom na čvrstoj fazi, pri čemu je korištena AccuBond ODS - C18 kolona. Pročišćeni uzorci analizirani su pomoću HPLC/MS sustava. Optimiranje je provedeno smanjivanjem količine otapala za eluaciju, čime je skraćeno trajanje metode. Metoda se pokazala uspješnom za detekciju 11 sulfonamida. Dokazana je linearnost, ponovljivost i ponovljivost mjerenja. Utvrđena je granica detekcije od 20 µg/kg. Dobivena iskorištenja pokazala su da je metoda dobra, točna i jednostavna. Analiza 33 uzorka meda pokazala je da su svi ispitani uzorci u skladu s propisima.

Abstract (english)

The sulfonamides are stable chemotherapeutics with wide range effect that are used in beekeeping for prevention and treatment of bee diseases. Their presence in honey in concentration above limit value is the potential danger to the human health. The limit value for sulfonamides in Croatia is up to 100 µg/kg. The aim of this work was optimation of a method for sulfonamides determination. The samples were cleaned by solid phase extraction on AccuBond ODS – C18 column. The cleaned samples were analysed by HPLC/MS method. The optimation was carried out by reducing an amount of solvent used for eluation, which shortened the procedure time. The method was efficient for detection of 11 sulfonamides. The linearity, repeatability and repeatability of measurement were determined. The established detection limit was 20 µg/kg. The parameters of efficiency proved that the method is fast, accurate and simple. The analysis of 33 honey samples has shown that all investigated samples satisfy the regulations.
