Abstract

Nesteroidni protuupalni lijekovi (NSAID, eng. nonsteroidal anti-inflammatory drug) intenzivno se istraţuju posljednjih desetljeća obzirom na njihov antitumorski potencijal te smanjenje rizika za razvoj i napredak malignih bolesti njihovom primjenom. Cilj ovog rada je ispitati in vitro učinak 25 različitih NSAID-ova na humanim staničnim linijama te antitumorski potencijal i mehanizam djelovanja onih s najizraţenijim citotoksičnim učinkom. Ispitani NSAID-i imaju potencijal kao antitumorski lijekovi obzirom na njihov jači antiproliferativni učinak na tumorske stanice, MCF-7, HepG2 i HeLa, nego na HEK293T stanice porijeklom iz zdravog tkiva, za koje su izračunate najveće vrijednosti IC50. Mogući mehanizmi djelovanja NSAID-a uključuju indukciju apoptoze, koja je značajnija kod tumorskih stanica nego kod normalnih stanica, te indukciju zastoja staničnog ciklusa u G0/G1 fazi odgovornu za antiproliferacijski učinak u tumorskim staničnim linijama.

Abstract (english)

Nonsteroidal anti-inflammatory drugs (NSAID) have been intensively investigated in the last decades in view of their anticancer potential and reducing the risk of developing and progressing malignant diseases by their application. The aim of this paper is to examine the in vitro effect of 25 different NSAIDs on human cell lines and the anticancer potential and mechanism of action of those with the most potent cytotoxic effect. Studied NSAIDs have potential as antitumor drugs due to their stronger antiproliferative effect on tumor cells, MCF-7, HepG2 and HeLa than on HEK293T cells derived from healthy tissue that have the highest calculated IC50 values. Possible mechanisms of NSAIDs action include induction of apoptosis, which is more significant in tumor cells than in normal cells, and induction of cell cycle arrest in the G0/G1 stage responsible for the antiproliferative effect in tumor cell lines.
