Abstract

Uljana repica i njeno ulje izvor su mnogih biološki aktivnih spojeva, osobito fenolnih spojeva za koje je potvrđeno da imaju antioksidacijsko, antimutageno i antitumorsko djelovanje. Iz sirovog ulja izoliran je 4-vinil-2,6-dimetoksifenol ili kanolol, a iz jestivog njegov dimer, fenilindan. U ovom radu ispitana je biološka aktivnost navedenih spojeva određivanjem antioksidacijskog kapaciteta ORAC metodom te in vitro testom citotoksičnosti na humanim tumorskim staničnim linijama (MCF-7, HeLa) te na normalnoj staničnoj liniji sisavaca (HEK293T). Ispitano je mogu li kanolol i fenilindan imati zaštitni učinak na stanice u kojima je izavan oksidativni stres. Rezultati istraživanja pokazali su citotoksičan učinak kanolola i fenilindana na sve tri stanične linije, s tim da fenilindan ima jači citotoksični učinak. Također, fenilindan ima veći antioksidacijski potencijal nego kanolol. Nije zabilježen zaštitni učinak kanolola i fenilindana na MCF-7, HeLa i HEK293T stanice u kojima je potaknut oksidativni stres.

Abstract (english)

Rapeseed and its oil are the source of many biologically active compounds, especially phenolic compounds that have been confirmed to have antioxidant, antimutagenic and anticancer activity. From crude oil is isolated 4-vinyl-2,6-dimethoksiphenol or canolol and from edible oil its dimer, phenylindan. In this study, we tested the biological activity of these compounds by determination of the antioxidant capacity by ORAC method and in vitro cytotoxicity in human tumor cell lines (MCF-7, HeLa) and in normal mammalian cell line (HEK). We examined whether canolol and phenylindane have a protective effect on the cells with induced oxidative stress. The results showed a cytotoxic effect of canololand phenylindane on all three cell lines, however phenylindan has a stronger cytotoxic effect. Also, phenylindane has a greater antioxidant potential than canolol. There had been no record of protective effect of canolol and phenylindane on the cells with induced oxidative stress.
