Rezistencija transformiranih sojeva kvasca s pojačanom ekspresijom odabranih gena na inhibitore rasta

Abstract

Jedan od pravaca razvoja u svrhu zadovoljavanja stalno rastućih potreba za energijom je proizvodnja biogoriva odnosno bioetanola iz otpadnih sirovina poljoprivrede, šumarstva i drvoprerađivačke industrije. Ove lignocelulozne sirovine su izvor jednostavnih šećera koji se fermentiraju u etanol. U ovim bioprocesima, zbog svojih mnogobrojnih prednosti, radni mikroorganizam je kvasac Saccharomyces cerevisiae. Međutim, spojevi koje kvasac može fermentirati, iz lignoceluloznih sirovina se dobivaju predobradom sirovine, tijekom koje nastaju i inhibitori rasta i fermentacije, kao što su octena i levulinska kiselina te 2-furaldehid. Budući da ovi spojevi uvelike otežavaju proizvodnju bioetanola, mnoga istraživanja su usmjerena prema konstrukciji novih sojeva kvasca, otpornijih na inhibitore rasta i fermentacije. Zbog toga je u ovom radu istražen utjecaj ekspresije osam gena na otpornost soja kvasca, izoliranog Sladorane d.o.o. Županja, prema pojedinim inhibitorima rasta. Dobiveni rezultati ukazuju na zaključak da pojačana ekspresija gena YAP1 i CTA1 doprinosi otpornosti kvasca na levulinsku kiselinu i 2-furaldehid te da na otpornost kvasca prema 2-furaldehidu pozitivno utječe i pojačana ekspresija gena ATR1 i GSH1.

Abstract (english)

In order to meet the constantly growing energy needs, one of directions in the production of biofuels or bioethanol is using waste materials of agriculture, forestry and wood processing industry. In these bioprocesses, due to its many advantages, the producer microorganism is the yeast Saccharomyces cerevisiae. Although lignocellulosic materials are the source of simple sugars that are fermented into ethanol, the compounds that the yeast can ferment are obtained by pre-treatment during which growth and fermentation inhibitors, such as acetic and levulinic acid and 2-furaldehyde, are also formed. Since these compounds greatly impede bioethanol production, different studies have focused on the construction of new yeast strains that are more resistant to growth and fermentation inhibitors. Therefore, the influence of the overexpression of eight genes on the resistance of the yeast strain isolated from Sladorana d.o.o. Županja, to individual growth inhibitors, was investigated in this paper. The obtained results suggest that overexpression of YAP1 and CTA1 genes contributes to yeast resistance to levulinic acid and 2-furaldehyde and that yeast resistance to 2-furaldehyde is enhanced by overexpression of ATR1 and GSH1 genes.
