Abstract

Brojna epidemiološka ispitivanja ukazuju na potencijalne pozitivne učinke kemijskih spojeva iz komine gožđa na ljudsko zdravlje. Posljednjih godina za njihovu ekstrakciju, kao alternativa konvencionalnim, predlažu se zelena eutektična otapala. U ovom radu ekstrakti komine grožđa pripremljeni su uz pomoć zakiseljenog etanola (GPEtOH) te kolin-klorid:citratne kiseline (GPChCit). Nadalje, pripremljenim ekstraktima ispitana je biološka aktivnost na dvijema staničnim linijama (HeLa i HepG2), spektrofotometrijskom metodom određen je udio ukupnih polifenolnih spojeva, a ORAC metodom određen je antioksidacijski kapacitet ekstrakata. Ekstrakt komine grožđa pripravljen uz pomoć eutektičnog otapala imao je veći antioksidacijski kapacitet i sadržaj polifenola od onog ekstrahiranog zakiseljenim etanolom. Ispitivanjem antiproliferativnog djelovanja in vitro na stanicama uočeno je inhibitorno djelovanje oba ekstrakta, koje je izraženije na HeLa stanice. Nije zapažena značajna razlika u djelovanju GPEtOH i GPChCit ekstrakta na rast HeLa i HepG2 stanica.

Abstract (english)

Numerous epidemiological studies indicate that chemical compounds isolated from grape pomace have potential positive effects on human health. Lately, as an alternative method for their extraction, green deep eutectic solvents have been proposed. In this study, grape pomace extracts were prepared using acidified ethanol (GPEtOH) and choline-chloride: citric acid (GPChCit). Furthermore, the biological activity of the extracts was tested on two cell lines (HeLa and HepG2), total polyphenols were analyzed spectrophotometrically and the antioxidant capacity was determined by the ORAC method. The grape pomace extract prepared by eutectic solvent have higher antioxidative capacity and higher amount of polyphenols than the one extracted by acidified ethanol. Investigation of antiproliferative activity in vitro on the cell lines showed the inhibitory effect for the both extracts, which was more pronounced in HeLa cells. There was no significant difference in the effect of GPEtOH and GPChCit extract on HeLa and HepG2 cell growth.
