Abstract

Ljubičaste nesumporne bakterije izolirane iz raznih staništa zanimljive su s ekološke strane i ekonomske strane u održivim procesima proizvodnje vodika kao biogoriva, s medicinske strane za sintezu biokemikalija kao aminolevulinska kiselina, za proizvodnju biopolimera (PHB) koji ne zagađuju okoliš i biomase te sintezu specifičnih spojeva kao karotenoida i pigmenata. Svrha održivog razvoja bioprocesa je korištenje obnovljivih izvora sirovina, primjerice lignoceluloznih sirovina. Cilj ovog rada bio je analizirati kinetiku rasta, potrošnju supstrata i sintezu fotosintetskih pigmenata ljubičaste nesumporne bakterije Rhodovulum adriaticum DSM 2781 tijekom rasta na podlogama koje sadrže glukozu i ksilozu kao izvor ugljika, te rast na hidrolizatima lignoceluloznih sirovina dobivenim kiselinskom predobradom u visokotlačnom reaktoru. Rezultati su pokazali da je najviši prinos biomase tijekom rasta na glukozi, a za sintezu pigmenata je najbolje koristiti podlogu s glukozom i ksilozom kao izvorom ugljika. Također je pokazano da je bakterija sposobna rasti na tretiranim i netretiranim hidrolizatima lignoceluloznih sirovina.

Abstract (english)

Purple non-sulfur bacteria isolated from different habitats are interesting from ecologic and economic point in sustainable processes for production of hydrogen as biofuel, in medicine for synthesis biochemicals such as aminolevulinic acid, for production of eco-friendly biopolymers (PHB) and biomass along with synthesis of specific compounds such as carotenoids and pigments. Purpose of sustainable development of bioprocess is usage of renewable sources, for example lignocellulosic materials. Aim of this thesis was analysis of growth kinetics, substrate utilization and synthesis of photosynthetic pigments in purple non-sulfur bacteria Rhodovulum adriaticum DSM 2781 during growth on media containing glucose and xylose as carbon source and analysis of growth on lignocellulosic hydrolysates obtained by acidic pretreatment in high-pressure reactor. Results have shown that highest yield of biomass was during growth on glucose and the best medium for pigment synthesis is mixture of glucose and xylose. It has been also shown that bacteria is capable of growing on treated and non-treated hydrolysates of lignocellulosic materials.
