Abstract

Oksidacijski stres je poremećaj ravnoteže između slobodnih radikala u organizmu te mehanizama kojima organizam neutralizira slobodne radikale. Mogući su endogeni i egzogeni izvori slobodnih radikala. Endogeni nastaju procesima metabolizma energije u organizmu, dok egzogeni nastaju kao posljedica unosa različitih ksenobiotika u organizam. Poliklorirani bifenili su aromatski ugljikovodici koji sadrže klorove supstituente na bifenilnom prstenu. Spadaju u skupinu ksenobiotika koja uzrokuje nastanak reaktivnih kisikovih vrsta, stoga mogu izazvati mutagenezu stanice, oštećenja DNA i stanica te apoptozu. Mehanizmi kojima organizam neutralizira slobodne radikale obuhvaćaju endogene i egzogene mehanizme, uključujući enzimske i neenzimske. U ovom radu provedeno je preliminarno ispitivanje utjecaja resveratrola na oksidacijski stres izazvan kongenerima PCB-77 i PCB-153 te kombinacijom navedenih kongenera na CHO-K1 staničnu liniju. Resveratrol nije pokazao protektivan antioksidacijski učinak prilikom predinkubacije CHO-K1 stanica s PCB-77, PCB-153 ni kombincijom tih kongenera.

Abstract (english)

Oxidative stress is defined as imbalance between production of free radicals in the organism and mechanism of their neutralization. The source of free radicals can be endogenous and exogenous. Endogenous free radicals are nusproducts of metabolism of energy, while egzogenous free radicals can be produced as consequence of xenobiotic intake. Polychlorinated biphenyls are aromatic hydrocarbons that contain chlorine supstituents in biphenyl ring. PCBs belong to a group of xenobiotics that induces the generation of reactive oxygen species which can lead to mutagenesis, cell and DNA damage and apoptosis. Free radical neutralization mehanisms can be endogenous and egzogenous mechanism, inclouding enzymatic and nonenzymatic. This preliminary thesis examines influence of resveratrol pretreatmant of CHO-K1 cell line on oxidative stress incuded by PCB-77 and PCB-153 congeners and the combination of this congeners. Resveratrol showed no protective antioxidative effect in preincubated CHO-K1 cells after exposure to PCB-77 and PCB-153 congener, as with a combination of these congeners.
