Abstract

Rast prisutnosti antibiotske rezistencije u 21. stoljeću zahtijeva strože analitičke kontrole prehrambenih proizvoda, a samim time i razvoj novih analitičkih metoda. Cilj ovog istraživanja bio je odrediti validacijske postupke u određivanju streptomicina i dihidrostreptomicina u medu primjenom UHPLC-MS/MS metode. Određivani validacijski parametri bili su: specifičnost, linearnost, ponovljivost, unutarlaboratorijska reproducibilnost, preciznost, mjerna nesigurnost, granične koncentracije i robusnost. Metoda se pokazala brzom, jednostavnom, te su rezultati validacije zadovoljili sve kriterije zakonskog okvira donesenog uredbom Europske komisije 657/2002. S obzirom na dobivene rezultate, metoda je prikladna za određivanje streptomicina i dihidrostreptomicina u medu.

Abstract (english)

Increase of antibiotic resistance in the 21st century demands more severe analytical control of food products, and hence the development of new analytical methods. The aim of this study was to determine validation procedures for the determination of streptomycin and dihydrostreptomycin in honey using the UHPLC-MS/MS method. Determined validation parameters were: specificity, linearity, repeatability, intra-laboratory reproducibility, precision, measurement uncertainty, limit of detection and quantification, and robustness. The method had proven to be quick and simple, and the results of the validation met all the criteria given by the European Commission Regulation 657/2002. Considering the acquired data, this method is suitable for the determination of streptomycin and dihydrostreptomycin in honey.
