Optimizacija uzgoja virusa sličnog varijantu QX u svrhu razvoja cjepiva ptičjeg infektivnog bronhitisa

Abstract

Virus ptičjeg infektivnog bronhitisa uzrokuje oboljenje peradi koje se najčešće manifestira poteškoćama u disanju, dok ponekad dolazi do nastanka lezija u bubrezima. U tom razdoblju perad gubi na težini i ne proizvodi kvalitetna jaja dopuštena za upotrebu. S obzirom na činjenicu da se virus lako prenosi i mutira, potrebno je učestalo razvijati nova cjepiva koja bi spriječila širenje sojeva virusa ptičjeg infektivnog bronhitisa do nezahvaćenih područja. Optimizacija uzgoja takvog živog cjepiva, soja virusa sličnog varijantu QX ispitana je pomoću metodologije dizajna eksperimenta. Korišten je potpuni faktorijalni dizajn u kojem su tri faktora ispitana na dvije razine, pri gornjoj i donjoj granici trenutnog uzgoja. Ispitani su faktori starosti embrija u kojima se uzgaja virus, početnog inokuluma kojim se embrij inficira i trajanja inkubacije, odnosno umnažanja virusa u embriju. Statističkom obradom rezultata pokusa predviđen je utjecaj pojedinog faktora te utjecaj međudjelovanja dva faktora i sva tri faktora na količinu virusa u potencijalnom cjepivu. Količina virusa je određena postupkom titracije virusa na primarnoj kulturi stanica bubrega pilećih embrija te izražena u obliku logaritma recipročnog razrjeđenja doze koja izaziva 50% promjena na inficiranoj staničnoj kulturi, log TCID50 mL-1. Tijekom uzgoja virusa sličnog varijantu QX statistički je najznačajniji utjecaj trajanja inkubacije virusa u embriju.

Abstract (english)

The avian infectious bronchitis virus causes a disease in poultry that is most commonly manifested by respiratory difficulties, while sometimes lesions in the kidneys can occur. During this period, the poultry loses weight and produces no quality eggs allowed for use. Since the virus is easily transmitted and mutated, the outbreak is prevented by the use of a viral vaccine. Optimization of cultivation of such live vaccine, variant QX-like was tested using the experimental design methodology. The full factorial design of experiments studied the effects of three factors, each at two levels, the upper and lower limit of current virus cultivation. Factors of interest are the age of the embryos in which the virus is grown, the initial inoculum by which the embryo is infected, and the duration of virus incubation. Statistical analysis of the results predicted the effects of a single factor as well as the effects of the interaction between two and all three factors on the final amount of virus in a potential vaccine. The amount of virus was determined by the virus titration method on cells of the primary culture of chicken embryo kidney cells (CEK cells) and expressed as the logarithm of reciprocal dose dilution causing 50% change of infected cells, log TCID50 mL-1. The factor of the duration of incubation time is statistically most significant.
