Abstract

Infekcije urinarnog trakta česta su pojava, osobito u ženskoj populaciji. Za liječenje infekcija urinarnog trakta koriste se pretežito antibiotici, no zbog pojave rezistencije često se pribjegava alternativnim metodama liječenja poput raznih biljnih pripravaka. Najčešće se koristi uvin-H čaja čijim se glavnim aktivnim sastojkom smatra arbutin, odnosno njegov metabolit hidrokinon. Hidrokinon uspješno eliminira brojne bakterije, poput E. coli, koje su najčešći uzrok infekcija urinarnog trakta. Cilj ovog rada bio je istražiti citotoksični i prooksidativni/antioksidativni učinak hidrokinona na stanice mokraćnog mjehura čovjeka T24. Također, istražen je i utjecaj hidrokinona na promjene u proteomu istim stanicama. Stanice karcinoma mokraćnog mjehura čovjeka su tretirane različitim koncentracijama hidrokinona tijekom 2 i 4 h u uvjetima bez oporavka i s 24-satnim oporavkom u hranjivom mediju. U ovom radu, dokazano je da je citotoksični učinak hidrokinona dozno ovisan. Isto tako, hidrokinon narušava integritet stanične membrane tretiranih stanica smanjujući zastupljenost proteina odgovornih za održavanje integriteta stanice. Oporavkom stanica u hranjivom mediju tijekom 24 h dolazi do obnove integriteta stanice.

Abstract (english)

Urinary tract infections are a common occurrence, especially in the female population. Antibiotics are mostly used to treat urinary tract infections, but due to the onset of resistance, alternative treatment methods such as various herbal preparations are often used. The most commonly used is Uva Ursi tea, whose the main active ingredient is hydroquinone, the metabolite of arbutine. Hydroquinone successfully eliminates many bacteria, such as E. coli, which are the most common cause of urinary tract infections. The aim of this study was to investigate the cytotoxic and prooxidative/antioxidative effects of hydroquinone on human urinary bladder cancer T24 cell line. The effect of hydroquinone on proteome of T24 cells was also investigated. The human urinary bladder cancer cells were treated with different concentrations of hydroquinone for 2 and 4 h under non-recovery conditions and with 24-hour recovery in culture medium. In this graduate thesis, the cytotoxic effect of hydroquinone has been shown to be dose-dependent. Likewise, hydroquinone impairs the cell membrane integrity of treated cells by reducing the amount of proteins responsible for maintaining cell integrity. Recovery of cells in the culture medium within 24 h results in the restoration of cell integrity.
