Abstract

Ptičji infektivni bronhitis je bolest koja uzrokuje probleme u peradarskoj industriji diljem svijeta. Bolest napada respiratorni sustav pilića čime izaziva velike ekonomske gubitke u peradarskoj industriji. Uzročnik bolesti je virus infektivnog bronhitisa koji je podložan čestim mutacijama i genomskim rekombinacijama. Zbog širokog spektra različitih serotipova i genotipova virusa potrebno je razvijati nova cjepiva. Optimizacija uzgoja živog cjepiva, glavnog alata u kontroli širenja infektivnog bronhitisa, soja virusa sličnog varijantu 2, ispitana je metodom dizajna eksperimenta. U istraživanje su bila uključena tri različita čimbenika: starost embrija u SPF jajima, veličina inokuluma i trajanje inkubacije. Količina virusa izražava se kao logaritam recipročnog razrjeđenja doze koja izaziva 50% promjena na inficiranoj staničnoj kulturi. Zajednički učinak primijenjenog titra inokuluma i trajanja inkubacije je imao najveći utjecaj na postignuti titar virusa sličnog varijantu 2.

Abstract (english)

The avian infectious bronchitis is disease that causes problems in poultry industry. A disease affecting the respiratory system of chickens and is associated with huge economic losses in poultry industry. Avian infectious bronchitis is caused by infectious bronchitis virus with great ability for mutation and genomic recombination. Because of many different serotypes and genotypes of virus the development of new vaccines is necessary. Optimization of variant 2-like virus cultivation as live vaccine, the most important method for controlling infectious bronchitis, was performed by the experimental design methodology. The experiments were performed by studying the effects of three different factors: the age of the embryos, the initial inoculum and the duration of virus incubation. The amount of virus was expressed as the logarithm of reciprocal dose dilution causing 50% change of infected cells. The greatest effect on the variant 2-like virus titer was achieved by the combined effect of the inoculum titer and incubation time.
