Abstract

Pod pojmom genetička transformacija podrazumijeva se laboratorijski postupak unošenja DNA u neku stanicu što rezultira promjenom genotipa i fenotipa transformirane stanice. Ovaj postupak se koristi u genetičkom inženjerstvu, a od iznimnog je značaja u biotehnologiji jer omogućava konstrukciju sojeva koji se koriste kao radni organizmi u biotehnološkim procesima. U ovom radu diskutirane su prednosti i mane metoda koje se najčešće koriste za transformaciju kvasaca kao što su metoda protoplastiranja, metoda pomoću litijevog acetata, elektroporacija, metoda pomoću staklenih kuglica, biolistička transformacija te transformacija pomoću bakterije Agrobacterium tumefaciens. Osim toga dane su najvažnije karakteristike vektora (molekula DNA) koje se koriste za ekspresiju heterolognih proteina i modifikaciju kvaščevog genoma te karakteristike pojedinih nekonvencionalnih kvasaca i primjeri njihove primjene u biotehnologiji.

Abstract (english)

The term genetic transformation refers to a laboratory procedure of introducing DNA into a cell, which results in a change of the genotype and phenotype of the transformed cell. This procedure has been used in genetic engineering, and is of great importance in biotechnology because it allows the construction of strains, which are used as working organisms in biotechnological processes. This paper discusses the advantages and disadvantages of the commonly used yeast transformation methods such as spheroplast method, lithium acetate method, electroporation, glass beads method, biolistic transformation and Agrobacterium tumefaciens mediated transformation. In addition, the most important characteristics of vectors (DNA molecules) used for the expression of heterologous proteins and modification of the yeast genome are given, as well as the characteristics of several unconventional yeasts and examples of their application in biotechnology.
